It is important to bear in mind that oxygen present in the atmosphere absorbs significantly below 200 nm and nitrogen absorbance assumes significance below 190 nm. Absorbance measurements made below 200 nm should be conducted with oxygen free nitrogen purging of sample compartment. However, for measurements below 185 nm it is best to maintain vacuum in the sample chamber.

- In case the sample needs incubation at a high temperature, care must be taken to avoid temperatures too hot for the cuvette.

Absorbance or transmission determinations shine spectrometer light directly through the cuvette to measure the amount of light absorbed or transmitted through the sample. In this measurement configuration, the opposite-side cuvette walls must be optically "clear" to let light pass unimpeded through the sample. The other two sides of the cuvette are often "frosted" to allow handling of the cell without dirtying the optical surfaces of the other two "measurement" sides of the cuvette. Historically, reusable quartz volume of a cuvette cuvettes were required for measurements in the ultraviolet range, because glass and most plastics absorb ultraviolet light, creating interference. Today there are disposable plastic cuvettes made of specialized plastics that are transparent to ultraviolet light. Glass, plastic and quartz cuvettes are all suitable for measurements made at longer wavelengths, such as in the visible light range.

On customer’s request they can be replaced with crystal quartz, sapphire or silicon windows. IR quartz has a usable wavelength range of 220 to 3,500 nm. It is more resistant to chemical attack from the sample solution than other types designed for fluorescence measurements.

Selection of a specific cell type will limit for example the appropriate temperature range, based on the selection of the dispersant and cell type. The manufacturing process requires each cuvette and cell to be assembled by direct fusion without the use of adhesives. This ensures resistance to chemical attack and extremes of temperature. Each cuvette and cell displays high transmittance values with performance features extending the spectrum from 190nm nm depending upon the chosen transmission material.
